Gimeracil (CAS: 103766-25-2) is an antitumor agent. Gimeracil is a potent inhibitor of dihydropyrimidine dehydrogenase (DPD), which inhibits the early step in homologous recombination for double strand breaks repair. Antimicrobials and antitumors for research and experimental use.
Gimeracil is a pyridi derivative with antitumor activity. Gimeracil enhances the antitumor activity of fluoropyrimidines by competitively and reversibly inhibiting the enzyme dihydropyrimidine dehydrogenase causing decreased degradation of the fluoropyrimidines.
Gimeracil has been administered concomitantly with 5-fluorouracil in a chemotherapy regimen termed S-1 to block 5-fluorouracil degradation, thus prolonging circulating 5-fluorouracil concentrations. Furthermore, at 1 mM Gimeracil has been shown to inhibit homologous recombination in the repair of DNA double strand breaks, which can sensitize cancer cells to radiotherapy.
